Summary

Nail fungus is a common and contagious infectious disease that starts as white or yellow spots under the nail tips. As the infection increases, the color of the nail changes, becomes thicker and breaks occur. They seriously impair quality of life and can cause pain. The treatment takes quite a long time and medication is required for a long time. Nail fungal diseases are also defined as "onychomycosis" and 50% of nail diseases are onychomycosis. One type of treatment is topical nail polishes. In topical therapy, the antifungal agent must penetrate between the layers of the nail plate at a minimum inhibitory concentration or higher to be effective.

Cyclopirox Olamine is a hydroxypyridine antifungal agent structurally different from common imidazole derivatives and other antifungal agents. Unlike other antifungal agents, it does not affect sterol biosynthesis. Studies on the mechanism of action have shown that the active substance of Ciclopirox acts by inhibiting the active transport and accumulation of macromolecular synthesis precursors into the cell. Cyclopirox showed fungistatic and fungicidal activity in broad-spectrum in vitro tests involving dermatophytes, yeasts, actinomyces, dimorphic fungi, eumyces and other fungal genera. Its commercial product of 8% is used in the treatment of onychomycosis.

Bioadhesion is the situation in which two surfaces, at least one of which is of biological origin, come into contact for a certain period of time under the influence of interfacial forces. Bioadhesive systems can be defined as systems that can control the release of drugs, perform an effective treatment in the targeted area, ensure a healthy life and play a role in ensuring continuity. It stays in the application area for a long time and thus provides a more effective treatment.

Within the scope of the project, HA and HEMA-g-Acrylamide polymers, which are bioadhesive polymers, were thiolated with thiol (SH) groups in order to increase their bioadhesive properties, and new polymers were synthesized.

In this study, synthesized polymers were used for the first time for a gel formulation indicated for the treatment of nail fungus. With thiolated polymers, it is aimed to increase the bioadhesion of the active substance on the nail surface and to increase the amount of active substance passing through the nail.
