Production of Orally Disintegrating Flexible Tablets and Use in Personalized Dosing

Project no. 21514 · Status: Kapanmış (Yayın bekleniyor) · 2020

Summary

Orally dispersible tablets (ODT) are solid dosage forms that disintegrate within seconds upon contact with a small amount of liquid. ODTs are designed and manufactured as user-friendly pharmaceutical forms to increase patient compliance compared to other solid oral dosage forms. In addition to patient compliance, one of the important factors in treatment is that the required dose should be easily given to the patient according to the condition of the individual and the course of the disease. However, since personalized dosing is not suitable for the large-scale production of drugs, the required dose for each patient and disease cannot be directly obtained. In this study, orally dispersible, flexible tablets were produced with desloratadine, used in the prevention of allergic reactions. Thus, it is aimed to increase its solubility in water, make it suitable for personalized dosing, and to improve its absorption and bioavailability. Studies have been carried out to eliminate the need for special packaging or instructions for the use required for existing orally dispersible tablets by ensuring that the tablets are in a flexible structure. The electrospinning method is the process of spraying the liquids in the electric field and depositing them in the fiber structure on the grounded collector. As a result of the electrospinning studies, different doses of desloratadine loaded mats and tablets were obtained by changing the production time. It has been seen that the obtained mats can be used directly, as well as being converted into tablet form with simple tablet compressing apparatuses that we designed and produced with a 3D printer during the project. When compared to the commercial product containing the same active substance but with a fragile structure, it was observed that the fabricated flexible tablets were not breaking against the applied force, however, they quickly dispersed in contact with a liquid, and the active substance was dissolved in the media. It is thought that the project outputs have the potential to increase the effectiveness of the treatment processes of many diseases, thanks to the use of this production technique and the developed formulations as base carriers.
