Summary

Ovarian cancer is the deadliest cancer of the female genital tract despite great advances in both surgical and oncological treatments. Exosomes are vesicular vesicles that are released into the extracellular environment by all known cells, from endothelium to fibroblasts and cancer cells. While cancer stem cell-derived exosomes regulate the microenvironment for tumor formation, the therapeutic properties of exosomes secreted from mesenchymal stem cells are dominant. In this study, it was aimed to determine the effect of mesenchymal stem cells on ovarian cancer cell line and to reveal their relationship with senescence markers. In the study, exosomes obtained from ovarian cancer cell line OVCAR3 and wharton jelly derived stem cells (WJ-MKH) were used as cell lines. Trypan blue dye was applied to check the viability and number of cells. Cells were counted at 10x magnification under an inverted microscope (Olympus). WJ-MKHs were incubated in a humid environment at 37°C and 5% CO. The characterization of exosomes obtained from WJ-MKHs was examined by flow cytometric method by looking at the exosome markers CD63 and CD9 (Beckman Coulter). The Real Time Cell Analysis System -xCELLigence (RTCA-SP) system was used to determine the cytotoxic effect of exosome suspensions from WJ-MKHs in the OVCAR3 cell line. Abcam beta Galactosidase Detection Kit (ab102534) was used for X-Gal Staining. As a result of the study, while the IC50 values of OVCAR cells treated with WJ-MKHs could not be obtained at the 24th hour, the IC50 dose at the 48th hour was 37.69 and the IC50 dose at the 72nd hour was 1429.3. It has been shown that the cytotoxic effect of exosomes on cancer cells is greater than that of stem cells and its effectiveness continues even at 72 hours. As a result of staining, no staining difference was found between the groups. It has been observed that exosomes obtained from WJ-MKH are bioactive agents with high therapeutic capacity with many advantages to the cell cycle of ovarian cancer.
