Investigation of the Effects of Hematopoietic Stem Cells and Synthetic Cannabinoid Application on Leukemia Cancer Stem Cells on Leukomogenesis
Project no. 20212 · Status: Kapanmış · 2019
Summary
Although natural cannabis tetrahydrocannabinol (THC) obtained from the cannabis plant is known to have various therapeutic benefits, synthetic cannabinoids (SC) obtained with chemicals in the laboratory environment are effective in the central nervous system and cause changes in perception, mood, consciousness, motor functions, behaviors by changing the functions of the brain. are psychoactive substances. The 9-THC, the psychoactive component of cannabinoid, signals through cannabinoid receptors (CB) accompanied by the G protein. CB1 receptor is abundant in brain tissues; The CB2 receptor has been identified in the lymphatic system. Therefore, the CB2 receptor is thought to be related to the lymphatic system. Synthetic cannabinoid binds to CB1 with 100 times higher affinity than cannabis. Leukemias, one of the lymphatic system diseases, are caused by the uncontrolled proliferation of hematopoietic stem cells, which are the first discovered stem cells that affect the lymphatic system in the body. During the development of stem cells into terminally differentiated cells, expressive changes are also observed in the chromatin dynamics of genes involved in epigenetic regulation. Studies on stem cells and cannabinoids have mostly been done with the natural cannabinoid THC. Since synthetic cannabinoids are generally used in studies requiring clinical observation and cell culture studies are very limited, their effects on stem cells are unknown. In this study, the role of Hematopoietic Stem Cells, Leukemia Stem Cells, which are blood cell precursors, in the development of leukemia will be investigated using JWH-018, a synthetic cannabinoid product. In our study, the IC50 dose will be determined by culturing the synthetic cannabinoid under appropriate conditions, and apoptosis, genetic and epigenetic expression analyzes will be performed. Since there is no detailed study on stem cells with synthetic cannabinoids; Through these analyzes, data on the effect of synthetic cannabinoid on stem cells and its role in the development of leukomogenesis were obtained.
