Development of a new radiopharmaceutical containing donepezil radiolabeled with Ga-68
Project no. 32257 · Status: Kapanmış (Yayın bekleniyor) · 2024
Summary

Within the scope of the project, a new radiopharmaceutical for parenteral administration containing Donepezil radiolabeled with Ga-68 will be developed. In the pharmaceutical part of the radiopharmaceutical, nanostructured carrier systems containing Donepezil will be prepared, and radiolabeling studies will be conducted. Nano systems have advantages such as their biocompatibility in parenteral applications, ability to be prepared in different sizes, high stability, potential use as therapeutic and imaging agents, and ability to be targeted to desired regions. In the project, drug carrier systems containing Donepezil, lipid-based, with positive or negative zeta potential, and aimed to have a particle size smaller than 500 nm will be prepared. These formulations, due to their active ingredient and the properties mentioned above, when radiolabeled with Ga-68, will easily cross the blood-brain barrier (BBB), making their use advantageous in Alzheimer's diagnosis.

When previous studies are examined, studies on the use of nanostructured carrier systems for imaging Alzheimer's disease have been found (Calderón-Garcidueñas et al., 2019; Chibhabha et al., 2020; Oyarzún et al., 2021). In these studies, formulations were used for radiological imaging. For the diagnosis of Alzheimer's disease, Fluorine-18 and Carbon-11 have been used as radionuclides in PET imaging studies (Rabinovici et al., 2007; Uzuegbunam, Librizzi, & Yousefi, 2020). When searches were conducted in international databases and websites (Pubmed, Science Direct, etc.) with the keywords Donepezil, Alzheimer, Ga-68, nanostructured carrier systems, and imaging, no articles were found. The absence of studies in the literature on the use of Ga-68 labeled Donepezil-containing nanostructured carrier system radiopharmaceuticals for Alzheimer imaging demonstrates the originality and innovative aspect of the project. Additionally, since this study will be a doctoral thesis, it will also contribute to young researchers gaining experience in this field.
