Summary

Amaryllidaceae alkaloids are an important group of secondary metabolites that have the potential to be used in different diseases depending on the diversity of their biological activities. Leucojum, Amaryllidaceae family, is represented by two subspecies worldwide and only Leucojum aestivum grows naturally in our country. This project includes a detailed quality control and phytochemical study on Leucojum aestivum grown around Lake Eğirdir in Isparta. The amount of loss in drying, total ash, hydrochloric acid insoluble ash, sulphated ash were determined separately in the underground and aboveground parts of the plant, and the results were calculated as percentages.

The amount of lycorine and galantamine, two important alkaloids of the Amaryllidaceae family, were determined by using high performance liquid chromatography (HPLC) method in the samples prepared from aerial parts and bulbs of the plant. Gas chromatography-mass spectrometry (GC-MS) was used to determine the alkaloidal profile of aerial parts and bulbs. As a result, totally 20 different alkaloids were detected in the plant.

The fractions obtained by fractionation of the total alkaloidal extract prepared from the dried and powdered total plant material at room temperature by column chromatography, were purified by using methods such as preparative column chromatography, preparative thin layer chromatography and crystallization. The structures of the obtained pure compounds were elucidated by spectroscopic methods such as 1D NMR, 2D NMR and MS.

According to the data obtained from theisolation and structure elucidation studies, galanthamine, narwedine from the galanthamine subgroup; lycorine, hippamine from the lycorine subgroup; tazettine, 11-deoxytazettine from the pretazettine subgroup; and a phenantridine type alkaloid 5,6-dihydrbicolorine were determined.

Anticancer activity study was performed on the estrogen-dependent breast cancer cell line MCF7 cell line by using MTT and LDH infiltration methods with these isolated compounds. The effects of the compounds on cell viability and LDH activity were determined.

Keywords: Leucojum aestivum L., Quality Control, Biological Activity
