ISOLATION OF CAROTENOID FROM HALOPHILIC BACTERIA ISOLATED IN IZMIR ÇAMALTI SALTERN AND DETERMINATION OF PRODUCTION POTENTIALS
Project no. 444 · Status: Kapanmış · 2017
Summary
Carotenoids considered as yellow-orange, red fat-soluble pigments and are usually composed of 40 carbons, found dominantly in algae, prokaryotes, and plants. Carotenoids have a variety of uses in the fields of biotechnology, cosmetic industry, and food technology. Çamaltı saltern is Turkey's largest salt lake and has great importance in terms of economic and ecological. it has the conditions for the production of halophilic bacteria which is important in biotechnology. In this study, 10 halophilic bacterial strains (SC1-10) producing carotenoid from Çamaltı Saltern were isolated using selective Marine agar medium. According to 16S rDNA gene sequence analysis, it was determined that the strains of bacteria belong as following: Bacillus safensis (SC4), Marinobacter aquaticus (SC2), Halomonas fontilapidosi (SC1, SC5), Halobacillus locisalis (SC7, SC8, SC9), Salicola salis (SC6), Halobacillus halofilus (SC10). From isolated strains, carotenoids were extracted by methanol / acetone method. After the extraction, carenenoids were determined by thin layer chromatography and then spectrophotometrically pre-screened at 300nm-800nm wavelengths. As a result of this pre-scan, SC1, SC2, SC3, SC4, SC5, SC6 and SC8 isolates were elected to further carotenoid analysis. The HPLC method was used for the detailed analysis and determination of caretenoids. HPLC analysis revealed that lycopene (SC1, SC3, SC5, SC2), ß-carotene (SC1, SC3, SC4), all-trans-lutein (SC2), 13-cis-ß-carotene (SC4), 9-cis-ß-carotene (SC4), apo-8-carotene (SC8) molecules were found to be produced by the isolates. Some carotenoid peaks were obtained which did not match the standards and literature data.
