BRAIN CANCER AND NEURODEGENERATIVE DISEASES TARGETED NOVEL AGENTS: 131I-BAICALEIN AND 131I-BAICALEIN-PLGA

Project no. 20178 · Status: Kapanmış (Yayın bekleniyor) · 2018

Summary

There is an important increase in neurological diseases. Researches on the development of brain agents and determination of their potential remain important due to the limitations of low specificity, tumor heterogeneity, side effects, drug resistance, blood-brain barrier as well as the unknown etiopathogenesis of diseases. Additionally, because of their natural properties and biological effects, the plant originated agents are under investigation for a long time and recently, they are preferred on diagnosis/therapy of diseases instead of synthetic compounds. In literature, Baicalein (BA) isolated from plant roots of Scutellaria Baicalensis Georgi has many pharmacological features such as anti-cancer, anti-diabetic, anti-oxidant, cardioprotective and neuroprotective. It has been observed that BA inhibits tumor metastasis and improves drug resistance of various drugs. In this thesis project, radioiodination and encapsulation studies of BA have been performed in order to develop targeted theranostic agents. According to characterization studies, it’s seen that PLGA encapsulated BA nanoparticles (BA-PLGA) were approximately 105 nm in size and have spherical morphology. Radioiodination was carried out by using Iodine-131 (131I) which is used for both diagnosis and therapy, thus radiolabeled BA and BA-PLGA (131I-BA and 131I-BA-PLGA) were obtained. According to quality control studies, radioiodination yield of BA and BA-PLGA were 100 % and 96 ± 0,70 %, respectively. In order to evaluate the potential that 131I-BA and 131I-BA-PLGA agents may have in neurooncology and neurodegenerative processes, the data obtained through in vitro cell culture studies on U87-MG and Daoy cell lines may have an important role in developing novel brain agents.
