The determination of Candida species in tissue samples of patients with intraoral premalignant and malignant lesions by DNA analysis and comparison with healthy tissue

Project no. 367 · Status: Kapanmış · 2017

Summary

Background: Aim of this retrospective study is to determine level of Candida albicans, Candida kruseii, Candida parapsilosis, Candida tropicalis, Candida glabrata in samples of patients with premalignant, malignant or benign lesions. Methods: Samples biopsied due to suspicion of premalignant or malignant lesions were included. They were divided into 4 groups: healthy (n=20), dysplasia (n=20), carcinoma in-situ (n=20) and squamous cell carcinoma (SCC) (n=20). Following DNA extraction, DNA samples were examined for quantity and quality. Quantitative measurements were performed using Real-Time PCR (LightCycler 96, Germany), standard curves were calculated. Data was analyzed using Mann-Whitney and Wilcoxon Signed Ranks tests (P <0.05). Results: C. parapsilosis (104 DNA/50 mg) was the most common Candida species among healthy and dysplasia groups. Level of C. tropicalis and C. kruseii (103 DNA/50 mg) was higher in carcinoma in-situ samples, C. kruseii (104 DNA/50 mg) was higher among SCC. Presence of C. albicans was not detected in any of the samples. No significant difference regarding level of different Candida species (P>0,05) were observed. When level of Candida was examined within each group, amount of C. glabrata (103 DNA/50 mg), C. parapsilosis (104 DNA/50 mg), C. tropicalis-C. kruseii (103 DNA/50 mg), C. kruseii (103 DNA/50 mg) was higher among healthy, dysplasia, carcinoma in-situ and SCC groups respectively. No significant differences were observed regarding level of Candida species within each group (P>0.05). Conclusion: The quantitative evaluation of Candida in healthy, dysplasia, carcinoma in-situ and squamous cell carcinoma samples using Real-Time PCR revealed similar levels of Candida among all groups.
