Development of Analytical Methods for Detection of Illegal Substances in Sweat and Comparison of Sweat Collection Methods Efficiency
Project no. 372 · Status: Kapanmış · 2018
Summary
The increasing abuse of legal and illegal substances has increased the number of analysis studies to detect these substances in the world. In forensic toxicological analyzes, blood and urine biological materials are widely used, sweat is considered as an alternative biological material. Sweat; secreted from the cells on the surface of the skin. Sweat has a slightly acidic nature, transparent, clear, has a narrow physiological range that are secreted to maintain constant body temperature, cumulative hypotonic bio-liquid for illegal and abusive substances. There are two different approaches, short-term (instantaneous) and long-term, for pad applications for sweat illegal substance analysis. The aim of this study was to investigate the effectiveness of long-term sweat patches and their differences with the short-term sweat collection materials (Drug wipe 5A, cosmetic pads, swabs) adopted under the 3001 - Initial R & D Projects Support Program. Within the scope of this project; a practical, effective, reliable analytical method for long-term detection of cannabis (?9-Tetrahydrocannabinol), morphine (MOR), cocaine (COC), benzoylecgonine (BE), amphetamine (AMP), methamphetamine (MET), 3,4-methylenedioxyamphetamine (MDA), 3,4-methylenedioxymethamphetamine (MDMA) was developed. An analytical method was developed for synthetic sweat samples using liquid-liquid extraction (LLE) technique. As a result of all solid phase extraction studies, repeatability and high recovery results could not be obtained (62-67%). Screening experiments were performed for 7 of the factors affecting LLE (sample volume, sample pH, ultrasonic bath time, ultrasonic bath temperature, LLE shaking time, derivatization time, derivatization temperature) and the results were statistically evaluated. 7 of 3 the most effective factors (sample volume, ultrasonic bath temperature, derivatization time) were used for the central composite design. The analytical method, whose chemometric optimum conditions were determined, was validated by the parameters in the international guidelines. The obtained data from the comparison of sweat collection methods can be used to create national production pads/swabs in the future were provided. The obtained data, were shared with the institutions and persons from the justice and health related departments, who is working for antidrug effort.
