Development of Nanostructured Lipid Carrier Systems Containing Lapatinib Ditosylate Targeted to Breast Cancer and In Vitro Evaluation of Targeting Efficiency
Project no. 31900 · Status: Kapanmış (Yayın bekleniyor) · 2023
Summary

Early diagnosis and effective treatment of cancer, which is the leading cause of death globally, significantly reduces the cost, duration and risk of mortality of treatment. Since radiological imaging is based on morphological changes, it is not sufficient for imaging the disease in the early stages. However, nuclear medicine imaging allows early diagnosis since it is based on the detection of physiological changes.

Since breast cancer is the most common type of cancer in women, early diagnosis of breast cancer is of great importance. Human epidermal growth factor receptor 2 (HER2) is observed to be excessively increased in 30% of breast cancer cases. This feature makes the HER2 receptor a possible target for targeting breast tumor tissue.

Compared to HER2-negative breast cancer, HER2-positive cancers tend to be poorly differentiated and high-grade, which generally leads to lower patient survival rates. HER2 plays a crucial role in the survival of cancer cells, making it a therapeutic target for many drugs, including small molecule tyrosine kinase inhibitors. Lapatinib ditosylate is an FDA-approved tyrosine kinase inhibitor used in the treatment of HER2-positive metastatic breast cancer.

The project plans to prepare nanostructured lipid carrier systems containing lapatinib and label them with 99mTc radionuclide. In this way, it is aimed to develop a new radiopharmaceutical that can be used for diagnostic purposes in nuclear oncology patients with high uptake in tumor tissue and low uptake in non-target tissue with molecular targeting. The usability of the radiopharmaceutical to be developed for diagnostic purposes will be evaluated with in vitro cell uptake studies.
