DEVELOPMENT OF POLYMERIC NANOMATERIAL SPECIFIC FOR CANCER TYPES AND ITS APPLICATIONS
Project no. 1239 · Status: Kapanmış · 2017
Summary
Early detection of cancer is vital to successful follow-up and survival of patients and therefore sensitive and specific methods are required. Analysis of biomarkers in biological fluids is one of the methods used to determine the disease. Mucin 1 is a type I transmembrane protein with a dense glycosylated extracellular domain extending from the cell surface to 200-500 nm. While expression of MUC1 in healthy epithelial cells is limited to the apical surface, MUC1 expression may occur from all surfaces due to loss of polarity in cancer cells. The expression of MUC1 in malignant adenocarcinomas is at least 10 times more, which makes it an perfect tumor marker for cancer diagnosis and treatment. In this study, nanopolymers developed by lectin affinity technique for detection of MUC1 glycoprotein were used as bioactive layer on electrochemical biosensor electrode surface. Nanopolymers were synthesized using mini emulsion polymerization method and derivatized with IMEO and ConA lectin. Advanced characterization studies of nanopolymers were performed. The operating conditions of the sensor system have been optimized. Biosensor validation studies were performed by determining analytical measurement characteristics. As part of the study, real sample blood serum was analyzed and this new method compared with a commercially available medical diagnostic kit. The new generation nanopolymeric material has been shown to be an affordable, sensitive, reliable and rapid device.
