IDENTIFICATION AND CHARACTERIZATION OF POTATO VIRUS Y (PVY) STRAINS IN SOME SOLANACEOUS PLANTS
Project no. 883 · Status: Kapanmış · 2018
Summary
In this project, the status of PVY strains in potato, tomato and pepper plants collected from İzmir Küçük Menderes Basin, where solanaceous plants are cultivated intensively and as polyculture, has been investigated. Also, it was aimed to analyze phylogenetically and characterize the PVY strains molecularly. In the research, 199 plant leaf samples including 65 potatoes, 79 peppers and 55 tomatoes were collected from surveys. Plant leaf samples from surveys were tested by serological (TAS-ELISA) and molecular (RT-PCR and multiplex RT-PCR) methods. 58,78% of the samples were found to be infected with various PVY strains. The most common PVY strains were PVYN:O with a rate of 67,7% in potato leaf samples, PVYNTN with a rate of 64% in tomato leaf samples and PVYC with a rate of 10% in pepper samples. As a result of phylogenetic analysis of the nucleotide and amino acid sequences of PVY strains detected in the solanaceous plants, the similarity rates with each other and with world isolates were determined. Accordingly, the two most similar isolates were PVYNTN/Potato and PVYNTN/Tomato isolates with a rate of 98,31%. Nucleotide similarity rates of PVYNTN/Potato- PVYN:O/Potato, PVYNTN/Potato-PVYN:O/Tomato, PVYN:O/Potato-PVYN:O/Tomato, PVYNTN/Tomato-PVYN:O/Potato and PVYNTN/Tomato-PVYN:O/Tomato isolates were 93,26%; 92,19%; 98,10%; 89,16% and 88,94% respectively. In the phylogenetic tree drawn by aligning the nucleotide sequences of the detected PVY isolates with each other, PVYC/Pepper isolate took place in a different branch separated from other tomato and potato isolates. In consequence of comparing the nucleotide sequences of PVY isolates with world isolates, PVYN:O/Potato isolate with 99,85% similar PVYPotato Egypt isolate, PVYNTN/Potato isolate with 99,77% similar PVYNTN/Pepper Polonya isolate and PVYO/Potato isolate with 98,87 similar PVYO/Potato England and South Africa isolates took place in closer phylogenetic groups. On the other hand, PVYNTN/Tomato isolate with 99,02% similar PVYNTN(a)/Potato ABD and PVYN:O/Tomato isolate with 12 world isolates displayed a nearby phylogenetic branching. Lastly, PVYC/Pepper isolate have been in close phylogenetic relationship with 93,99% similar PVYTomato Spain isolate to itself.
