Evaluation of Bilayered Osteochondral Scaffold in Treatment of Cartilage Defects - Experimental Study in Rabbits
Project no. 22275 · Status: Kapanmış (Yayın bekleniyor) · 2020
Summary

In this study, a novel bi-layered scaffold that developed by a local company, was evaluated to repair osteochondral defects in rabbit knees. This novel scaffold’s upper (cartilage) layer consists of polyglicolic acid (PGA), hyaluronic acid (HA) and lower (bone) layer consists of β-tricalcium phosphate (β-TCP). The purpose of this study was to evaluate the efficacy of this novel scaffold, combined with or without mesenchymal stem cells (MSCs) to repair osteochondral defects in rabbit knees.

Thirty osteochondral defects were created in rabbits left femoral troklea. 10 defects were treated with scaffold combined with MSCs (group A), another 10 defects were treated with cell-free scaffolds (group B), the others were untreated (group C). At 12 weeks macroscopic and histopathologic evaluation were done.

The mean International Cartilage Repair Society (ICRS) macroscobic scores were 4.95 for group A, 6.16 for group B, and 8.25 for group C. The mean Oswesty Arthroscobic Scores (OAS) were 1.65 for group A, 3.39 for group B and 6.05 for gruop C. The macroscobic scores were significantly higher in group C than group A for ICRS scores and group A and group B for OAS scores (p<0.001, p<0.000, p<0.022). Macroscopic findings were also supported by histopathological findings.

In summary, our findings suggest that in a rabbit model, the novel osteochondral scaffold is not effective for repairing full-thickness osteochondral defects with or without MSCs. Further researches are needed for improving this novel osteochondral scaffold.
