Summary
Cancer is the second cause of death in the world and in our country. This disease gains the ability for cells to uncontrollably with changed features physically and biochemically. Surgery, chemotherapy, radiotherapy are most commonly used for cancer treatment. Natural products are biologically active substances which have been used for treatment for many years. In recent years, scientists search for new agents isolated from natural sources, because of the limitations such as drug resistance and high toxicity. In this project, we loaded saponin molecules to silica nanoparticles that we synthesized and characterized. But after lots of testing, loading capacity couldn’t reached high efficiancity. Limitations of time and sources, silica nanoparticle method was removed and the biological activities of saponins were investigated. We investigated the cytotoxic and apoptotic effects of davisianoside B and aristatoside C molecules, which were isolated from respectively Cephalaria davisiana and Cephalaria aristata, on lung cancer cell lines. A549 (lung carcinoma), HTB-177 (large cell carcinoma), CRL-5807 (bronchioalveolar non-small cell carcinoma), CRL 5826 (squamous cell adenocarcinoma; mesothelioma), and CCD-34Lu (normal lung fibroblast) were cultured. Cytotoxic effects on cells, were determined by MTT test. For the apoptosis activity, cells analysed with FITC conjugated Annexin V by flow cytometry. It is shown that saponins of davisianoside B and aristatoside C have a potential of cytotoxic effect on lung cancer cells, and at low concentrations have a greater cytotoxic activity than the commercial chemotherapeutic drug doxorubicin. Additionally, molecules have apoptotic activity potential dose-dependent manner on lung cancer cells.
