Summary

This in-vitro study aimed to compare the effects of calcium silicate-based Biodentin and glass ionomer-cement (Fuji IX) on remineralization in artificial dentin lesions and comparatively evaluate the microleakage when used as restorative material. In the first part of the study four standart cavities in each tooth (1x1x1 mm) were prepared on thirty-four dentin sections obtained from extracted sound human third molars to detect remineralization. One cavity was covered with acid-resistant nail polish (Gr 1, positive control). Specimens were soaked in demineralization solution at 37 º C for 96 hours to induce artificial caries lesions. Biodentin (Gr 2) and Glass ionomer cement (GIC, Gr 3) were applied to the other cavities, fourth cavity was unrestored (Gr 4, negative control). Then they were immersed in simulated body fluid (SBF) for twenty-one days. Vertical sections were obtained perpendicular to the cavities. In order to detect remineralization, SEM images of the cement-dentin interface were evaluated and Ca/P values in dentin adjacent to the restoration were determined using SEM-EDX. The data were statistically analyzed using Bonferroni test. For microleakage testing, thirty nine non-carious third molars were randomly assigned to three groups (n:13): Gr A: Biodentin, Gr B: GIC , Gr C; Composite (positive control). The flat dentin surface was exposed by taking a horizontal section from the occlusal third of teeth and an occlusal cavity of 4x2x2 mm was prepared on the dentin surfaces. They were than restored with the materials as manufacturers’ instructions. After thermocycling the samples were covered with nail polish, keeping the surfaces 1 mm away from the restoration edges. They were stained with basic fuchsin solution, sectioned in buccolingual direction and measured for microleakage. The staining at the restoration-dentin interface was scored in the stereomicroscope by two examiners. The data were evaluated statistically using Kruskal-Wallis test (p <0.05). According to the findings of the remineralization study, Ca/P values were significantly higher in Gr 2 compared to the Gr 3. It was determined that Ca / P values in Gr 2 and Gr 3 were significantly higher than (Gr 4). When Ca / P values in Gr 2 and Gr 1 were compared, it was significantly higher in the Biodentin group. No significant difference was found between Ca / P values in Gr 3 and Gr1. When analysing the data for microleakage, there was a significant difference between Gr A and Gr B (p> 0.1). No statistical difference in microleakage was found between Gr A and Gr C, while Gr B exhibited higher microleakage than Gr C. According to the mean rank, it was determined that the average microleakage values of the Biodentin group were the lowest compared to the other groups. Within the limitations of this in-vitro study it is concluded that both the remineralization ability and the sealing performance of Biodentin were higher than glass ionomer cement. It was thought that Biodentin can be used for restorative purposes. However, long-term clinical studies should be conducted to evaluate the success of the material.
