Project no. 23613 · Status: Kapanmış (Yayın bekleniyor) · 2022

Summary

Lung cancer is the most frequently diagnosed type of cancer in the world and has the highest number of cancer-related deaths. The prognosis of lung cancer is relatively poor compared to other types of cancer, and approximately 75% of patients can be diagnosed in advanced stages. The leading cause of lung cancer is the use of cigarettes and tobacco products with a rate of 90%, while the other 10% may be caused by factors such as radon gas, asbestos, exposure to air pollution and chronic infections. In addition to all these, lung cancer is a type of cancer that can be inherited. Lung cancer is divided into two histological classes, which grow and spread in different ways. These; Small Cell Lung Carcinomas (SCLC) and Non-Small Cell Lung Carcinomas (NCLC). In the treatment of lung cancer, different treatment modalities are applied according to the homes of the disease. These are radiation, surgery, chemotherapy treatment or targeted therapies. Cisplatin is one of the most commonly used drugs in chemotherapy treatment in lung cancer. Cisplatin is a platinum-based anticancer drug known as Cisplatin or cis-diaminedichloroplatin (II). In addition to lung cancer, cisplatin is also used in the treatment of many different cancers such as bladder, head and neck cancer, and testicular cancer. Although the use of cisplatin is quite common, the development of resistance in the patients in the later stages of the treatment may cause the treatment to be limited. Recent studies show that the ER stress marker, Glucose Regulated Protein 78 (GRP78), is effective on drug resistance. The elucidation of the mechanism underlying the development of cisplatin resistance is very important for the treatment process and more studies are needed to elucidate this mechanism. The use of herbal active substances in cancer treatment is increasing day by day. The effect of Resveratrol, one of these active ingredients, on cisplatin-resistant lung cancer cells is not fully known. In this study, the effects of Resvaratrol on Cisplatin-resistant and sensitive Calu-1 cells were investigated by cytotoxicity study, and also its effects on GRP78 chaperone proteins in these cell lines were investigated by QPCR and apoptosis studies. According to the results obtained, the effect of Resveratrol on the development of Cisplatin resistance and whether it can contribute to a new treatment approach due to its effects on GRP78 gene expression has been examined.
