Interrelationships of CAPN-1 and CAPN-2 in breast cancer

Project no. 29588 · Status: Kapanmış · 2023

Summary

Although the structures of the two major calpain isoforms (calpain-1: CAPN-1; calpain-2: CAPN-2) expressed in all mammalian cells are quite similar, it has been reported that they can perform different/opposite functions depending on the situation. Since calpains are targeted in cancer treatments, it has become important to fully determine the role of CAPN-1 and CAPN-2 in cancer. Within the scope of a COST project completed by our group, it was observed that CAPN-2 protein level increased significantly when CAPN-1 expression was silenced in HCC1937 cells (human breast cancer). The absence of any study in the literature showing that CAPN-1 controls the protein level of CAPN-2 made this data very interesting and encouraged our group to conduct further studies. Thereupon, our searches in databases showed that CAPN-1 is high in breast cancer and CAPN-2 is low. These results indicate that CAPN-1 controls CAPN-2 and that they may play opposing roles in breast cancer progression. In our project proposal based on this, we aimed to examine the relationships between CAPN-1 and CAPN-2 in breast cancer cells, to reveal the mechanism of the control of CAPN-1 over CAPN-2, and to investigate the possible opposing roles of these proteases. The fact that there is no study focusing on the opposing roles of CAPN-1 and CAPN-2 in cancer and that no other data has been encountered showing that CAPN-1 regulates CAPN-2 makes this project unique. The data obtained from this project will draw attention to the roles of CAPN-1 and CAPN-2 in specific types of cancer and encourage scientists to conduct studies on this subject.
