Preparation of Solid Lipid Nanoparticle Formulations for the purpose of ING4 gene delivery and showing their effectiveness on different cancer cell cultures
Project no. 258 · Status: Kapanmış · 2016
Summary
Purpose of this Project is; preparation of DNA delivery systems that carrying ING4 gene sequence and show their effectiveness in cell culture after performing the characterization studies. ING4 is a protein that induces apoptosis by inhibiting growth in cancerous cells. It has been observed that the gene region encoding this protein in many cancer types has been deleted or protein synthesis decreased. Therefore, it is thought that ING4 gene transfer may decrease the reproduction and proliferation of cancer cells. In this study, solid lipid nanoparticles were prepared using the hot microemulsion method to prepare the DNA delivery system. First of all, different oil and surfactant mixtures were titrated with water to form ternary phase diagrams and W/O (water / oil) microemulsion zones were defined. Then, by making characterization studies for the formulations selected in these microemulsion regions, formulations with optimal particle size and zeta potential values were determined to complex with plasmid DNA. Stability studies were performed by loading DNA to selected formulations, and their effectiveness in cancerous and healthy cell cultures was shown. In this way, gene therapy is aimed for the ING4 gene which has been deleted or reduced in cancer cells.
