The effect of Palosuran on Lung cancaer cell

Project no. 105 · Status: Kapanmış · 2017

Summary

Urotensin-II is a somatostatin-like cyclic small peptide molecule characterized by its potent vasoconstrictor activity. It has been described from many tumor tissues. Perhaps it will be possible to use it in human cancers as an alternative prognostic marker. It is thought to play a role in angiogenesis, cell proliferation and tumor biology. Urotensin II is thought to have these effects by stimulating autocrine and paracrine growth through promitogenic patway PKC (Protein Kinase C) - ERK1 / 2 (Extracellular Signal Regulated Kinase) and p38 MAPK (Mitogen Activated Protein Kinase). Lung cancer is the most common cause of cancer-related deaths globally for both sexes. Advances in the diagnosis and treatment of lung cancer in recent years have made significant contributions to the survival and lifespan of patients. Similarly, with the intensive molecular studies carried out, the approach to lung cancer and treatment principles have been completely renewed. The aim of our study is to demonstrate the effects of urotensin II receptor antagonist palosuran on the promitogenic signaling pathway in lung cancer cell lines, to demonstrate the cytotoxic activity of palosuran in vitro and to determine IC50 doses. In the long term, it is aimed to demonstrate the possible synergistic cytotoxic / apoptotic effects of palosuran with cisplatin, to test this combination in vivo based on the promising results to be obtained from the study, and to form a basis for a new combination therapy in the clinic.
