Synthesis, Characterization and In Vitro Efficacy Determination of Ixabepilone Loaded Folic Acid Bound Alginate Chitosan Nanoparticle Blends
Project no. 23821 · Status: Kapanmış · 2022
Summary

The incidence and mortality rates of cancer types are increasing day by day due to environmental factors and biological factors including genetic disorders in tumour suppressor genes, genetic predispositions and familial inherited cancers. Among cancer types, breast cancer is one of the leading health problems that increase in terms of incidence in the world and is known as the most common type of cancer in women. According to new data published by the International Agency for Research on Cancer (GLOBOCAN), approximately 2.3 million (11.7%) new cases of breast cancer in 2020, surpassing the most commonly diagnosed lung cancer. Even if there are improvements in early detected breast cancer patients, the difficulties in the treatment of metastatic patients and the low survival in these patients cause breast cancer to be the most serious cause of high mortality among women and the most common type of cancer. Ixabepilon, used in the treatment of metastatic breast cancer, is a non-taxane tubule polymerising agent and is effective in patients resistant to taxane group drugs. Patients with breast cancer and metastatic breast cancer may have nerve damage (neuropathy) due to the use of cancer drugs. Ixabepilone has similar side effects, making its use restrictive. Within the scope of this proposed project, it is aimed to prepare IXP-CSFA (IXP-CSFA), IXP-ALGFA (IXP-ALGFA) and IXP-CSFA/IXP-ALGFA blend nanoparticle systems to be used in the treatment of breast cancer and to determine their in-vitro efficacy. For this purpose, folic acid targeted chitosan alginate nanoparticle system is envisaged to provide effective and safe treatment of ixabepilone active substance at lower doses, as well as to reduce side effects and increase bioavailability. For this purpose, it is envisaged that the folic acid-targeted chitosan alginate nanoparticle system will provide effective and safe treatment at lower doses of the active ingredient ixabepilone, reduce its side effects and increase its bioavailability. Within the scope of the project, folic acid was bound to alginate (FA-ALG) and chitosan (FA-CS) separately in the first stage. Ixabepilone-loaded FA-ALG nanoparticles and ixabepilone-loaded FA-CS nanoparticles were prepared and then blends of drug-loaded nanoparticles were obtained. Quality control studies, drug release (profile) kinetics, in vitro cell cytotoxic activity of the prepared IXP/FA-CS and IXP/FA ALG nanoparticle forms and IXP/FACS//IXP/FA-ALG blends on MDA-MB-231 breast cancer cell lines expressing folate receptor and MCF-7 breast cancer cell lines not expressing folate receptor were determined, and their therapeutic potential was revealed.
