EFFECT OF SEAWEED EXTRACT APPLIED TO GWOWTH MEDIA ON MYCELIUM GROWTH OF OYSTER MUSHROOM

Project no. 21266 · Status: Kapanmış · 2019

Summary

This study was carried out to determine the effect of Ecklonia maxima type seaweed extract (DY) applied to growth media on mycelium growth of oyster mushroom (Pleurotus ostreatus) in January, 2020. In the experiment, 2 different growth media [malt ekstract agar (MEA) and potato decrose agar (PDA)] were used. After the prepared MEA (48 g L-1) and PDA (39 g L-1) medium was autoclaved at 121 ° C for 20 minutes, half was separated and 2% E. maxima seaweed extract was added. The experiment consisted of 4 topics: MEA, PDA, MEA + DY and PDA + DY. While the prepared environments were warm, they were filled in petri dishes under UV light in a sterile cabinet and left to cool. After disinfection of rootstock oyster mushrooms with alcohol, clones were taken and placed on the medium in petri dishes and petri dishes were closed with parafilm. 15 petri dishes from each treatment were prepared and the petri dishes were placed in the incubator at 24 °C. Micelle lengths (cm day-1) and micelle weights (g petri-1) were measured regularly. Among the fattening media used, PDA was found to be more hopeful to provide winding and form heavier micelles in a shorter time than MEA. DY application increased micelle development and shortened the winding time. DY performed better in PDA environment. As a result of the research, PDA + DY application in beech mushroom development is suggested because it increases micelle development and speed and decreases development time.
