ANALYSIS OF SURVIVAL OF FAT GRAFTS USED IN PLASTIC SURGERY WITH BIOINFORMATICAL METHODS

Project no. 22173 · Status: Kapanmış (Yayın bekleniyor) · 2020

Summary

Using fat tissue as a graft is a frequently used method in Plastic Surgery. However, the biggest limitation of this technique is that it requires re-injection of fat due to the resorption of the majority of the fat placed, and the etiology that leads to repetitive surgeries has still not been revealed.

One of the most important medical discoveries recently made is Platelet Rich Plasma (PRP). This method is the local application of growth factors such as VEGF and FGF, which are densely found in the granules of platelets, to affect the desired tissue. However, these studies are only observational studies on a statistical basis, without showing a cause-effect relationship. The aim of our study is to reveal the molecular etiology of fat tissue resorption and the mechanism of action of PRP in this phenomenon. Although many studies have been conducted on these subjects before, the fact that detailed and comprehensive research has not been conducted at the molecular level increases the original value of our study and distinguishes it from other studies.

A New Zealand rabbit will be used in the study. The right inguinal fat pad will be collected as the Sham (Blind) Group. Then, 5 cc of blood will be taken to obtain PRP. The remaining bilateral inguinal fat pads will be excised. Fat tissues will be obtained as nanografts and injected into the parascapular areas with 1 cc of PRP on the left and 1 cc of SF on the right, following weight and volume measurements. (Right parascapular = Control Group; left parascapular = Experimental Group). At the end of one month, the remaining tissues from the injected fat grafts will be excised. The obtained tissues will undergo transcriptome analysis using Next Generation DNA Sequencing. Expression differences determined by bioinformatics analyses will be compared for the three groups.

If the project is completed successfully, the information obtained will contribute to the fields of Medicine and Bioinformatics and will guide new projects to be carried out subsequently.
