Summary

Matrix metalloproteinases and tissue inhibitors play critical roles in the extracellular matrix. They also have important effects on training and sports performance. The aim of the study was investigate the levels of matrix metalloproteinase-9 (MMP9) and tissue inhibitor of metalloproteinases-1 (TIMP1) before and after acute maximal exercise and the role of MMP9 -1562 C/T and TIMP1 372 T/C polymorphisms in athletes and sedentary subjects. Athlete group includes 43 basketball, volleyball, handball players (age: 20,65±2,46) sedantery group includes 43 healthy men (age: 22,49±3,51). YOYO IR-1 was applied to determine the endurance levels of the participants. A blood test conducted before and after the test. MMP9, TIMP1, MMP9/TIMP1 ratio, blood lipids and lipoproteins [total cholesterol (TC), high and low density lipoprotein cholesterol (HDL-C ve LDL-C), triglyceride (TG)], hemogram, glucose, urea, creatinine, indicators of oxidative stress [total antioxidant status (TAS), total oxidant status (TOS), oxidative stres index (OSI)], indicators of muscle damage and inflammation [creatine kinase (CK), aspartate aminotransferase (AST), alanine aminotransferase (ALT), interleukin-6 (IL-6), C reactive protein (CRP)] were determined from satiety venous blood. Polymorphisms were analyzed from DNA samples obtained from peripheral blood. MMP9 and TIMP1 levels were found higher in both groups after the YOYO IR-1 test (exercise) (p<0,05). MMP9 -1562 C/T and TIMP1 372 T/C polymorphisms had no effect on MMP9 and TIMP1 levels. The effect of exercise, group and genotype interaction for MMP9 -1562 C/T polymorphism on AST was found statistically significant (p<0,05). The amount of AST increase in athletes with CT genotype was higher than sedentary subjects with CT genotype. TIMP1 372 T/C polymorphism had a significant effect on TK and LDL-K levels and individuals with C allele had higher TK and LDL-K levels than individuals with T allele (p<0,05). The effect of exercise and allele interaction for TIMP1 372 T/C polymorphism on LDL-K was found statistically significant (p<0,05). The amount of increase of LDL-K was higher in individuals with C allele than individuals with T allele. These findings suggest that acute exercise increase MMP9 and TIMP1 levels in male atheletes and sedantary subjects but MMP9 -1562 C/T ve TIMP1 372 T/C polymorphisms are not associated with this enzyme and its inhibitor.
