Özet

Abstract

Background: Macroalgae play a crucial role in aquatic ecosystems and are increasingly studied for their potential in cancer research. Enzymes, such as DNA topoisomerase, play a significant role in regulating defects in DNA repair mechanisms at the beginning of the cancer process. These enzymes generate transient single or double-chain breaks in the DNA and bind covalently to the DNA via tyrosine residues in the enzyme's active sites. The gap in the DNA is then repaired by rejoining the temporary break. Material and method: To achieve this, we will obtain extract fractions/biomolecules from C. crinita using the EtOH extraction method. We will then indicate the cytotoxic activity of the obtained fractions on healthy B lymphocytes, leukemia cell lines, and leukemia stem cells. To do this, we will perform a total and partition algal extraction. Then, treatment of the fractions to cell lines to determine their cytotoxic activity and identify candidate extracts with cytotoxic activity. Finally, The TOPO1 super spiral relaxation and TOPO2 decatenation activity of the candidate extracts and identify extracts that show TOPO1 or TOPO2 inhibitory activity at the lowest concentration. Results: To extract the algae material, the algae collected from the coasts were dried and processed to obtain the total EtOH extract. The C. crinita EtOH extract was suspended with water and partitioned with n-hexane, ethyl acetate (EtOAc), and n-butanol (BuOH). The cytotoxic effects of 1 total extract and 4 partitioned extracts were investigated and their IC50 doses were determined. The most effective extracts in the cytotoxicity assays were BuOH and EtOAc. The study was continued with the fractionation of BuOH and EtOAc extracts. Based on the cytotoxic data obtained, these two fractions were analyzed for their activities to inhibit the topoisomerase enzyme. Conclusion: When the inhibition effects of the fractions on topoisomerase enzymes were analyzed according to the data obtained, it was observed that the EtoAc and BuOH extracts showed TOPO1 and TOPO2 enzyme inhibition activity even at the lowest concentration of 0.02 mg/mL. These extracts show promise as a potential biomolecule. Keywords: Macroalgae, Topoisomerase, Cytotoxicity Activity
