Özet

Parkinson’s disease (PD) is a progressive neurodegenerative disorder primarily managed through pharmacological treatments aimed at symptom control. The mainstay of therapy includes oral administration of levodopa and dopamine receptor agonists. However, oral delivery faces significant challenges, including fluctuations in drug plasma levels and extensive hepatic metabolism, which can reduce therapeutic efficacy. Selegiline hydrochloride (SH), a monoamine oxidase-B (MAO-B) inhibitor, is widely used alone or in combination with levodopa, but its effectiveness is often compromised due to rapid first-pass metabolism. Emerging research highlights the potential of non-oral drug delivery systems to enhance therapeutic outcomes and enable personalized treatment strategies. This study explores the development of buccal formulations as a convenient and effective alternative for Parkinson’s disease therapy. Oromucosal films were systematically evaluated for key physicochemical and functional properties, including pH, weight, thickness, swelling behavior, disintegration time, drug content, tensile strength, mucoadhesion, in vitro drug release, cytotoxicity, neuroprotective effects, and stability. The selected selegiline hydrochloride oromucosal film exhibited favorable characteristics, including a smooth texture, uniform thickness and drug distribution, physiologically compatible surface pH, strong mucosal adhesion, controlled swelling behavior, and sustained drug release. These properties support its potential as a patient-friendly and efficient dosage form. The findings suggest that oromucosal films offer a promising platform for selegiline hydrochloride delivery, providing an innovative alternative to conventional therapies and warranting further clinical investigation.
