Özet
Macrophage migration inhibitory factor (MIF) is a pleiotrophic lymphocyte and macrophage cytokine; it is likely to play an important role in innate immunity. Despite the importance of MIF in innate and acquired immune responses, until recently, few mechanisms had been reported that could adequately explain MIF's functional contribution to inflammatory and autoimmune disease. To analyse the association of the −173 G/C polymorphism of the MIF gene genotype and allele frequencies with biliary atresia. Between February 2003 and November 2004, 12 patients (mean age 0.6 ± 0.4) diagnosed as biliary atresia. After informed consent, blood was collected and DNA was obtained. MIF 173 C/G polymorphism was detected using the PE ABI Prism 310 Sequence Detection System (SDS) for multicolor real-time or endpoint fluorogenic polymerase chain reaction detection. They also compared with children with chronic liver disease (n = 42) and a group of unrelated healthy controls (n = 133). Associations were assessed by using Fisher's exact test. MIF 173 gene genotype and allele distribution according to groups is summarized in table 1. Univariate analysis showed that MIF-173G/C polymorphism was significantly related to biliary atresia. When compared with the control group the frequency of the MIF-173 C allele was significantly higher in biliary atresia patients than chronic liver disease and healthy controls.(p = 0.04). Further analysis confirmed that MIF-173C allele was independent risk factor for biliary atresia (p = 0.03, OR = 5.1; 95%CI 1.3-20.36). Our results strongly suggest that −173G/C polymorphism of MIF gene might be associated with the susceptibility of biliary atresia.
