Özet
Stem segments with 3-5 leaf axills excised from the upper portion of the main stem of 2-year-old Araucaria heterophylla (Norfolk Island pine). Seedlings were produced from the ortotrophic buds of the concealed axillary meristems when cultured on a basal medium (BM) of half strengthed Murashige and Scoog (1962) inorganic salts, mineral salts of deFossard (1981), growth factors and 20 g/l sucrose and 6-5 g/l agar. Bud tubercules appeared after exposing to growth in basal medium with 1% active coal for 35 days and then subcultured. Liquid rooting medium were prepared with the addition of 10 μM IBA to the ¼ of salt compounds of MS medium. In this medium, buds were left at 22-25 oC for two weeks. Occurring plantlets were transferred into pods including perlid turbomoss (40/60) in a medium of 90-95 % humidity. Then the pods were left to the acclimatization. Floral buds from Odontoglossum grande (Tiger orchid) Lindl grown in the Botanical Garden were cut to be used as explant in January. A Vacin and Went (1949) liquid medium was prepared with the addition of 10 kDa chitosan (15 ppm) and 150 ml cocconut milk invitro using a sterilised filter. The medium was prepared at pH: 4.9 in two ways; the first was without agar while the second with 12 gr/l agar. After six weeks exposure to liquid medium, rooted protocorms were transferred to the solid medium. Every green protocorms gave 5-6 plantlets.
