Summary
The utility of microfabricated thick-film sensors for trace measurements of insulin was examined. The procedure relied on potentiometric stripping analysis (PSA) at screen-printed carbon electrodes, involving an adsorptive accumulation of insulin and the oxidation of the tyrosine residues of the adsorbed hormone. Different carbon inks, printing and activation conditions, and relevant variables of the stripping protocol (accumulation potential and time, stripping current) were assessed and optimized. The optimal procedure allowed convenient monitoring of trace insulin (down to the 20 nM level) in a physiological pH in connection to 2 min accumulation periods. ne high sensitivity was coupled to a good precision. The response is affected by interferences from electroactive species and co-adsorbing constituents, that would require proper attention prior to the realization of single-use insulin strips. Ultimately, we envision a dual glucose/insulin strip based on coupling common chronoamperometric biosensing of glucose with a highly sensitive stripping detection of insulin.
