Özet

Spinal cord injury (SCI) results in devastating neurological deficits due to the disruption of motor, autonomic, and sensory functions. In this study, we aimed to investigate the neuroregenerative effects of graphene in in vitro models of SCI, focusing on mechanical and chemical injury. NSC-34 motor neuron-like cells were employed in mechanical and chemical injury models to simulate SCI. Mechanical injury was induced using a scratch model, while chemical injury was achieved through exposure to alpha-kainic acid (α-KA). Graphene was applied at varying concentrations to assess its efficacy in promoting cell viability and scratch closure. Immunohistochemical staining was conducted to examine neurite outgrowth and neuronal marker expression. In the mechanical injury model, NSC-34 cells exhibited gradual migration and proliferation, leading to scratch closure within three days. Chemical injury with α-KA resulted in dose-dependent cytotoxicity, with an effective dose of 70 µM. Treatment with graphene significantly improved cell viability, accelerated scratch closure, and enhanced expression of neuronal markers. Our findings demonstrate the promising neuroregenerative and neuroprotective effects of graphene in in vitro models of SCI. Graphene holds potential as a therapeutic agent for SCI, offering opportunities for enhancing neural regeneration and improving patient outcomes.
