In this study, it was aimed to investigate the protective effects of Boldine alkaloid in a rotenone-induced toxicity model in SH-SY5Y cells. Although Boldine is known for its strong antioxidant and anti-inflammatory effects, its protective activity against rotenone was investigated for the first time.

Methods: In this study, rotenone were applied to the SH-SY5Y cell line for 24 hours and the LD50 dose of rotenone was determined as 200 nM. To determine protective activity, cells were pretreated with various doses of Boldine for 2 hours, followed by exposure to 200 nM rotenone for 24 hours. As a result of the applications, cell viability was determined by MTS test, and reproduction/colony formation abilities were determined by clonogenic test. Caspase-3 ELISA and Hoechst staining were performed to assess the type of cell death. One-way ANOVA was used for statistical analysis.

Results: Our results showed that 200 nM rotenone killed approximately half of the cell population (p<0.0001). It was observed that cell survival and colony formation ability increased significantly when boldine treatment was applied for 2 hours before rotenone (200nM) application (p<0.0001). In addition, pretreatment with boldine decreased rotenone-induced increased chromatin condensation and active caspase 3 levels (p<0.0001).

Conclusion: The results of our study revealed that rotenone decreased cell viability by increasing apoptotic cell death in neuronal cells. Boldine has an antiapoptotic effect, increased cell survival and colony formation ability against rotenone-induced neurotoxicity at low doses, and it was observed that this protective activity decreased as the dose increased. With this study, the protective efficacy of boldine was demonstrated for the first time in an in vitro model of Parkinson's induced with rotenone.
