Introduction: Epigenetics is changes in DNA that does not involve the genetic sequence but leads to changes in translation. Epigenetic modification of asthma genes needs to be clarified to discover therapeutic targets. The aim of this research was to identify the status of methylation as an epigenetic mechanism in the promoter region of e-cadherin (ECAD) gene in children with asthma. Methods: We enrolled children aged between five and 15 years that are diagnosed with atopic or nonatopic asthma in our Pediatric Allergy and Pulmonology department. Children who received inhaled or systemic steroids more than three weeks duration were excluded. Control group included non-asthmatic children who presented to Healthy Child department of the same institute. Age, sex, socioeconomic status of the family, total IgE and eosinophil count were recorded. 2 ml blood sample was obtained for DNA isolation with a standard protocol. Methylation specific PCR (MSP) was performed on these DNA samples to evaluate the status of methylation in the promoter region of the ECAD gene. Result: ECAD promoter gene methylation was evaluated in samples obtained from children with atopic asthma (n=20), nonatopic asthma (n=22) and nonasthmatic controls (n=20). Methylation status was not found to be significantly different betwen the groups (p=0.734). Conclusion: The results of our research did not demonstrate a significant difference in the methylation status of ECAD gene in asthmatic children. However, considering the role of epithelial barreir dysfunction and ecadherin in pathogenesis of asthma, further reseach to measure the extent of methylation of this gene may provide valuable information.
