The techniques involved in the study of toxin activity include the determination of sensitivity of tissue by electrolyte leakage preparation of radioactive labelled HS toxin and toxin purification, membrane and protein preparations, equilibrium dialysis with scintillation counts of 14C-HS, and lipid preparation. Toxin binding proteins have been found in membranes of non-hosts. Toxin activity is thought to occur on the plasmalemma, where it perturbs the membranes, releasing bound ATPase which lyses cells. A major protein, able to bind the toxin, is present on the plasmalemma, external to the cytoplasm. However, presence of other membrane components is apparently needed for toxin binding and symptom development.

Keywords: Bipolaris sacchari, helminthosporoside toxin, specific side proteins.
